摘要:
为了解不同普通栽培品种枇杷中FCA基因对花发育的影响,延长枇杷的销售时间,以花期相差一个月且开花数量差异较大的‘解放钟’及西班牙枇杷品种爱沙拉’两个枇杷品种的芽、花苞和花为材料,提取RNA并克隆获得‘解放钟’和‘爱沙拉’两个枇杷品种的EjFCA基因,将获得的EjFCA基因进行测序并做生物信息学分析。结果显示,‘解放钟’和‘爱沙拉’中EjFCA基因的CDS序列分别为1125 bp和1137 bp、分别编码374和378个氨基酸,二者的相似度高达98.15%。利用生物信息学软件分析表明两个枇杷EjFCA基因均含有1个RRM结构域和1个WW结构域,保守性较高,为GATA类转录因子;亚细胞定位预测显示EjFCA蛋白位于细胞核,与苹果和白梨的亲缘关系最近。对EjFCA在上述两个枇杷品种的表达分析显示EjFCA基因在两个枇杷品种的芽,花苞和花中均有表达,在‘解放钟’中呈“先上升,后下降”的趋势,花苞时期表达量最高,在盛花期略有下降,但在‘爱沙拉’枇杷的3个花期中表达量变化不大,且始终低于‘解放钟’。上述研究表明调控‘爱沙拉’枇杷花期的主要基因不是EjFCA,但其花芽的形成可能受EjFCA调控。(图8 参22)
Abstract:
The purpose of this study was to understand the effect of FCA gene on flower development in different cultivars of loquat . Buds, alabastrums and flowers of ‘ Jiefangzhong ’ and Spanish loquat variety ‘ M.Aixaza ’ were used as materials . There was a month difference in the flowering period between the two loquats. The EjFCA genes of two loquat were cloned by RNA extraction. The EjFCA genes were sequenced and analyzed by bioinformatics. The CDS sequences of EjFCA gene in ‘ Jiefangzhong ’ and ‘ M.Aixaza ’ were 1125 bp and 1137 bp, respectively, encoding 374 and 378 amino acids, respectively. The homology of two FCA genes was 98.15%. Bioinformatics analysis showed that the two loquat EjFCA genes contained one RRM domain and one WW domain, which were highly conservative. The predicted results by PlantTFDB indicated that EjFCA gene was a kind of GATA transcription factor. The prediction of subcellular localization showed that EjFCA protein was located in the nucleus and microsome, and had the closest genetic relationship with apple and pear. The expression of EjFCA in two loquat showed that EjFCA genes were expressed in buds, alabastrums and flowers of ‘ Jiefangzhong ’ and ‘ M.Aixaza ’ . The expression of EjFCA was the highest in the period of alabastrums, and decreased slightly at the full flowering stage. However, the expression of loquat in ‘ M.Aixaza ’ had little change during the three flowering stages, and was always lower than ‘ Jiefangzhong ’ . It is suggested that EjFCA was not the main gene in the regulation of flowering period of ‘ M.Aixaza ’ , but the formation of flower bud may be regulated by EjFCA.
