- STR是植物萜类吲哚生物碱（TIAs）生物合成的中心酶，它所催化的PS反应生成的异胡豆苷是TIAs的合成前体物。本研究从喜树基因数据库中挖掘筛选到14条注释为STR的候选基因，并通过生物信息学以及RT-qPCR方法，分析其编码蛋白的理化性质、结构特征、系统进化关系以及在喜树无菌幼苗不同时期和不同部位的基因表达情况。序列结构特征分析表明，CaSTR1、CaSTR2、CaSTR3、CaSTR7、CaSTR8、CaSTR11、CaSTR12、CaSTR13和CaSTR14含有五个典型的XXX#§保守区，且在第五个XXX#§区N端邻近区域存有特征的EVXE区。系统进化树分析表明，喜树CaSTR序列多与葡萄和棉花的SSL序列聚为一支，亲缘关系较近。基因相对表达分析结果表明，9条CaSTR候选基因序列在喜树幼苗组织中均有表达，且40 d时CaSTR1、CaSTR2、CaSTR3、CaSTR4、CaSTR7、CaSTR12、CaSTR13候选基因在叶中表达量最高，与喜树碱积累特征一致。由此判断CaSTR1、CaSTR2、CaSTR3、CaSTR7、CaSTR12、CaSTR13候选基因可能参与喜树碱的生物合成。本研究为后续喜树STR候选基因的功能研究提供理论依据。（图5 表3 参30）
- Abstract:
- STR is the central enzyme in the biosynthesis pathway of terpenoid indole alkaloids in a variety of plants . Strictosidine, which is produced by the PS reaction catalyzed by STR, is a crucial precursor for the synthesis of TIAs . In this study, 14 candidate genes annotated as STR were mined and screened from the genomic database of Camptotheca acuminata. Their protein physicochemical properties, structural features , phylogenetic relationship and gene expression patterns in different periods and different parts of C. acuminata seedlings were analyzed by bioinformatics and RT-qPCR. Sequence structure analysis showed that most of the CaSTR protein sequences contained five typical XXX#§ conserved regions , and a characteristic EVXE region was also observed in adjacent location s. Phylogenetic analysis showed that most of the CaSTR sequences and the SSL sequences from Vitis vinifera and Gossypium arboretum were clustered together and closely related . The relative gene expression analysis showed that nine CaSTR candidate genes were expressed in seedling tissues . The candidate genes of CaSTR1, CaSTR2, CaSTR3, CaSTR4, CaSTR7, CaSTR12 and CaSTR13 were most ly expressed in the leaves of 40 d, which is similar with the expression pattern of camptothecin. Thus we can conclude that CaSTR1, CaSTR2 , CaSTR3 , CaSTR7 , CaSTR12 , CaSTR13 candidate genes may be involved in the biosynthesis of camptothecin. This study laid a foundation for the functional characterization of STR candidate genes in C. acuminata.
