摘要:
葛根芩连汤是治疗肠道湿热证泄泻的经典方剂,为了解其疗效机理,以“高糖高脂+高温高湿+白酒+冰水”的方法制备肠道湿热证泄泻小鼠模型,运用葛根芩连汤进行治疗。治疗前后无菌采集小鼠肠道内容物和肠黏膜,测定分析样品中总微生物活度以及蔗糖酶、淀粉酶、乳糖酶和蛋白酶活性。结果表明,与正常组相比,模型动物肠道微生物活度显著升高(P < 0.01),肠道内容物中乳糖酶、蔗糖酶和淀粉酶活性升高而蛋白酶活性降低,肠黏膜中乳糖酶、蔗糖酶和淀粉酶活性显著降低(P < 0.05)而蛋白酶活性升高。葛根芩连汤治疗后,治疗组肠道微生物活度降低向正常组水平变化。除蛋白酶外,治疗组肠道内容物中乳糖酶、蔗糖酶、淀粉酶活性显著高于正常组(P < 0.05)而低于自愈组(P < 0.05),肠黏膜中乳糖酶、蔗糖酶和淀粉酶活性显著高于正常组(P < 0.05)和自愈组。本研究表明葛根芩连汤可能通过降低肠道湿热证泄泻小鼠肠道微生物活度,调节肠道内容物和肠黏膜中乳糖酶、蔗糖酶、淀粉酶和蛋白酶活性而发挥疗效。
Abstract:
Ge-gen-qin-lian decoction (GD) is a classical prescription of traditional Chinese medicine (TCM) to treat diarrhea with intestinal dampness-heat snydrome (IDHS), but the treatment mechanism is not very clear. Diarrhea model with IDHS was established with high fat and sugar diet, high humidity and temperature, gavage of white wine, administrated with icy water. Then GD was given to the diarrheal mice. Before and after treated with GD, intestinal contents and mucosae of mice were collected. The activities of microbe, sucrase, amylase, lactase and protease of samples were determined. The results showed that the intestinal microbial activity of the model group were increased significantly (P < 0.01) compared with the control group, the activities of lactase, sucrase and amylase in intestinal contents of the model mice were increased, while protease activity were decreased, however the activities of lactase, sucrase and amylase in intestinal mucosae of the model mice were significantly decreased (P < 0.05). After administration with GD, the microbial activity in the treatment group decreased toward the level of the control group. Except protease, the activities of lactase, sucrase and amylase in contents of the treatment group were significantly higher than that in the control group (P < 0.05), while significantly lower than that in the untreated group (P < 0.05). The activities of lactase, sucrase, and amylase in the mucosae of the treatment group were higher than that in the control (P < 0.05) and untreated mice. The results obtained herein indicate that GD can decrease the intestinal microbial activity, regulate the activities of lactase, sucrase, amylase and protease in intestine of mice, which might reveal the therapeutic mechanism of GD on treating diarrhea with IDHS.
