摘要:
黄精市场需求量不断增加,较低的自然繁殖速度难以满足人们的需求。通过组织培养技术,建立黄精快速繁殖技术体系,可以高效获得大量优质种苗在生产上应用。首次以四川主载品种大叶黄精地下根茎芽为外植体进行了组培快繁体系的初步研究。研究表明:外植体消毒先用0.5 g/L的多菌灵溶液预处理12 h,再用0.2% HgCl2灭菌6 + 6 min,污染率可得到有效控制;四川大叶黄精根茎芽最适宜的初代培养基为MS + 6-BA 0.5 mg/L + NAA 0.5 m g/L,萌发率达35%,长势健壮;不定芽分化培养基为MS + 6-BA 3.0 mg/L+ NAA 0.2 mg/L,平均不定芽诱导个数为4.33个;壮苗培养基为MS + 6-BA 1.5 mg/L + 2,4-D 0.5 m g/L + GA 3 0.5 g/L;生根培养基为1/2 MS + NAA 1.0 m g/L + IBA 0.2 m g/L + AC 0.2 g/L,生根率达89%。以大叶黄精的地下根茎芽为外植体可快速获得大量组培苗,其中细胞分裂素6-BA在大叶黄精生长过程中起着重要作用。(表5 参26 附图4)
Abstract:
The current reproduction rate of Polygonatum sibiricum is hard to meet the demand for Polygonatum which has been increasing rapidly . In order to provide high quality seedlings of Polygonatum kingianum Coll. et Hemsl. var. grandifolium D. M. Liu et W. Z. Zeng, var. nov. , predominant cultivars of Polygonatum sibiricum in Sichuan, we develop ed a rapid micro propagation system by taking it’s rhizome buds as explants. The results show that the contamination rate can be effectively controlled when the explants were pretreated with 0.5 mg/l carbendazim solution for 12 h, before being sterilized with 0.2% HgCl 2 for 6 min+6 min; the optimal medium for primary culture was MS+6-BA 0.5 m g/L +NAA 0.5 m g/L, germination rate was 35.0%; the optimal medium for adventitious bud induction was MS+6-BA 3.0 mg/L +NAA 0.2 m g/L; the optimal medium for cultivating strong seeding was MS+ 6-BA 1.5 mg/L + 2,4-D 0.5 m g/L +GA 3 0.5 m g/L, while the medium 1/2 MS+NAA 1 mg/L + IBA 0.2 m g/L + AC 0.2 g/L was suitable for the rooting and the rooting rate was 89%. A large number of tissue culture seedlings can be obtained quickly by using the underground rhizome buds of Polygonatum sibiricum as explants. Cytokinin 6-BA plays an important role in the growth of Polygonatum sibiricum.
