摘要:
生长素反应因子(Auxin Response Factors,ARFs)在植物生长发育过程中调节生长素响应基因表达发挥着重要的作用,为了解多花水仙ARF家族,基于转录组数据,通过NR、NT、Swiss-Prot和PFAM 4个数据库、NCBI网站和在线软件SMART的注释筛选,利用ExPASy-ProtParam tool、SOPMA、Prot Comp、MEME、MAGA和Heml软件进行氨基酸理化性质、蛋白二级结构、亚细胞定位、保守基序、系统进化树和小鳞茎萌发不同时期的表达模式分析。共筛选得到21个ARF家族基因,生物信息学分析12个全长基因蛋白长度在193-1096aa之间,预测的分子量为20.46-122.2kD,等电点为5.19-7.97,全部为不稳定的亲水蛋白,亚细胞定位预测均位于细胞核中。进化树和基序分析表明,21个NtARF蛋白分为3个大组,Glass Ⅲ进一步被分为3个亚组,分支相近的ARF转录因子具有相同或相近的基序。NtARF8、NtARF13、NtARF15、NtARF17和NtARF21都展现较高的表达模式,可能是通过正向调节生长素的含量来促进小鳞茎的萌发。本研究表明NtARF家族可能在多花水仙扦插繁殖过程中发挥着重要作用,结果可为将来深入研究多花水仙ARF基因家族的功能奠定基础。
Abstract:
Auxin Responsive Factors (AFRs) play an important role in regulating the expression of auxin-responsive genes during plant growth and development. To understand the ARF family of Narcissus, based on transcriptome data, t he NR, NT, Swiss-Prot, PFAM, NCBI sites and online software SMART were used at the same time to annotate and screen. ExPASy-ProtParam tool, SOPMA, Prot Comp, MEME, MAGA and Heml software were used to analyze the physical and chemical properties of amino acids, secondary structure of proteins, subcellular localization, conservative motifs, phylogenetic trees and expression patterns of bulblets at different stages of germination. The results showed that 21 ARF family genes were screened. Bioinformatics analysis showed that 12 full-length ARF gene proteins ranged from 193 to 1096aa, with the predicted molecular weight varied from 20.46 to 122.2 kD and the isoelectric point in the range of 5.19 to 7.97. All of them were unstable hydrophilic proteins, and subcellular localization predictions were located in the nucleus. Evolutionary tree and motifs analysis showed that 21 NtARF proteins were divided into three groups. The GlassⅢ could be further divided into three subgroups , and the similar branching ARF transcription factors had the same or similar motifs. NtARF8, NtARF13, NtARF15, NtARF17 and NtARF21 all showed higher expression patterns, which may promote the germination of small bulbs by positively regulating the content of auxin. The study indicated that NtARF family may play an important role in the cuttage propagation of Narcissus, which laid a foundation for further study on the function of ARF gene family of Narcissus.
