Abstract:
In order to improve the mixed storage quality of dry maize straw and cabbage wastes, the dynamic effect of Pichia anomala inoculation on the fermentation quality was studied. The experiment was conducted with an additive-free control group (ME group) and microbial treatment group (PA group) inoculated Pichia anomala. The two feedstocks were continuously stored and fermented at constant temperature for 170 days by the ensiling theory, and the sensory quality, chemical composition and fermentation quality were analyzed at intervals. The high-throughput sequencing technology of Illumina Miseq was used to investigate the dynamic changes of bacterial community diversity during mixed ensiling. The results showed that the content of dry matter, crude protein, water soluble carbohydrates and lactic acid in group PA increased significantly compared to group ME (P < 0.05). In contrast, the content of pH value, acid detergent fiber, neutral detergent fiber in group PA decreased significantly compared to group ME during storage for 170 days (P < 0.05). The ratio of lactic acid to acetic acid, the ratio of lactic acid to total organic acid were both improved by the inoculation of P. anomala, which showed that P. anomala improved ensilage fermentation quality. The results of bacterial community diversity showed that the dominant bacterium communities in group PA inoculated with Pichia anomala were the Firmicutes and Proteobacteria in phylum level, and Lactobacillus and Paralactobacillu were dominant in genus level. The abundance of spoilage bacteria such as Enterobacter, Pseudomonas and Flavobacterium decreased after inoculation of P. anomala, and Enterobacter disappears after storage of 130 d. The abundance of lactic acid bacteria in group PA always remain above 50%. In conclusion, the inoculation of P. anomala can improve the fermentation quality of dry maize straw and cabbage wastes, so that the dried straw can be stored for nearly half a year for its resource utilization.
