摘要:
肠道微生物与人体多项生理功能相关,是中药药效发挥的重要作用者,研究七味白术散对菌群失调腹泻小鼠肠道乳酸杆菌的影响,可以为其疗效机理提供依据. 通过灌胃硫酸庆大霉素和头孢拉定对小鼠进行肠道菌群失调造模,然后灌胃给药七味白术散全量传统汤药和50%量超微汤药对造模小鼠进行治疗,提取肠道微生物宏基因组DNA,用乳酸杆菌特异引物对肠道微生物的16S rDNA进行PCR扩增,通过Hha I、Rsa I、Sam I和Hind Ⅲ等酶切后进行ARDRA分析. 结果发现:正常组和七味白术散50%剂量超微汤药治疗组OTUs数大于七味白术散传统汤药治疗组,七味白术散作用下乳酸杆菌的多样性指数、均匀度指数及优势度指数分别为0.330、0.091和0.913,均有所恢复;聚类与主成分分析表明,七味白术散对肠道乳酸杆菌具有一定的调整作用,且超微七味白术散50%量汤药的治疗效果优于七味白术散传统汤药. 本研究结果说明七味白术散对菌群失调腹泻的疗效与调整肠道乳酸杆菌有关,七味白术散50%超微汤药的效果较之全量传统汤药的效果更好.
Abstract:
Gut microbes are closely associated with multiple human physiological functions, thus play a significant role in the pharmacological effects of traditional Chinese medicine. This study aimed to investigate the micro-ecological functions and their mechanism of ultra-micro powders of Qiweibaizhusan on diarrheal mice with dysbacteriosis. The intestinal dysbacteriosis in mice were induced by oral administration of gentamycin sulfate and cefradine capsules. After that the effects of traditional decoction of Qiweibaizhusan and ultra-micro powders of Qiweibaizhusan (with dose equivalent to approximately 50% that of traditional decoction of Qiweibaizhusan) were observed on the diarrheal mice. The intestinal microbial metagenomics were extracted and then PCR amplified using lactobacilli specific primers. The diversity of PCR products were analyzed with amplified rDNA restriction analysis (ARDRA). The results showed that the OTUs of lactobacilli were more in mice of normal group and ultra-micro powders Qiweibaizhusan group than in those of the traditional decoction Qiweibaizhusan group. The diversity, uniformity and dominance of lactobacillus were recovered by treatment with ultra-micro powders of Qiweibaizhusan and traditional decoction of Qiweibaizhusan. The results of cluster analysis and principal component analysis also showed that both ultra-micro powders of Qiweibaizhusan and traditional decoction of Qiweibaizhusan adjusted intestinal lactobacilli, with ultra-micro powders being more effective than traditional decoction of Qiweibaizhusan. The diversity of intestinal lactobacilli in the group of ultra-micro Qiweibaizhusan was comparable to that of the normal group, better than that in the traditional Qiweibaizhusan group and the model group. This study provides a scientific basis for the clinical application of ultra-micro powders of Qiweibaizhusan.
