摘要:
摘要 对小麦waxy蛋白的提取技术和电泳后胶片的银染方法进行了改良∶将无胚的半粒种子研磨成粉,用洗液浸泡2 h代替蒸馏水浸泡过夜,并且去掉了用洗液反复洗涤和离心的程序;电泳后胶片的染色采用快速银染法代替常用的银染法.改良后的单向十二烷基硫酸钠聚丙烯酰胺凝胶电泳方法(1DSDSPAGE)具有省时、方便、经济和有效的特点,可适用于大批小麦材料的waxy蛋白亚基的分析. 图1 参5
Abstract:
Abstract The methods for extraction of wheat waxy protein and silver staining of gel are improved. When extracting waxy protein, crushed seeds are dipped into washing buffer for 2 h rather than distilled water for overnight, moreover, washing granule repeatedly with washing buffer and microfuging are omitted; and gel is stained with rapid silver staining. The results show that the improved 1DSDSPAGE method for identification of wheat waxy protein is more convenient, rapid, cheap and available, especially for bulks of wheat. Fig 1, Ref 5
