独岛枝芽胞杆菌(Virgibacillus dokdonensis)1A00493分离自深海,对水稻细菌性病害水稻黄单胞菌和严重危害植物根部的南方根结线虫都具有较好的拮抗效果. 根据我国2012年最新公布的微生物农药毒理试验行业标准,通过小鼠急性经口毒性试验、急性经呼吸道毒性试验和急性注射毒性试验,评价V. dokdonensis 1A00493发酵液对哺乳动物的生物安全性. 对于3种染病途径的毒性评价分别采用灌胃、鼻腔滴注、静脉注射方式一次性给药,观察小鼠的临床表现并测定小鼠的体重和脏器系数. 结果显示:V. dokdonensis 1A00493发酵液各剂量不同给药方式的处理小鼠均未出现明显中毒症状及死亡现象,且对小鼠的生长发育无不良影响. 小鼠体重变化与对照相比较并没有显著性区别(P > 0.05),脏器系数的测定结果与对照组比较差异不显著(P > 0.05),各脏器解剖没有出现病变. 因此V. dokdonensis 1A00493发酵液对小鼠安全无毒副作用,可应用于农业生产中病害的防治. Antagonistic bacterium Virgibacillus dokdonensis 1A00493 isolated from deep sea has antimicrobial and nematicidal activity. It has good activity against Xanthomonas oryzae pv. oryzae pxo99 that causes the most severe bacterial diseases to rice and Meloidogyne incongnita that causes serious damage to crops. With an aim of laying foundation for safe application of this antagonist bacterium in the disease control during agricultural production, in this research we did mouse acute oral toxicity, acute respiratory toxicity and acute injected toxicity tests to evaluate the safety of V. dokdonensis 1A00493, according to the newly published Standard Toxicology Test of Microbial Pesticides in 2012. The mice were treated with the fermentation liquid of V. dokdonensis 1A00493 by single-dose intragastric, nasal drip or intravenous administration. Then the physical signs, body weight and visceral index were observed and recorded. The results showed no evident poisoning symptom or death in treated groups; no adverse effect on the growth and development of mice was observed. Neither the body weight of mice nor visceral index measured results showed significant differences from the control group (P > 0.05). The viscera showed no pathological changes in gross dissection. It indicated that the fermentation liquid of V. dokdonensis 1A00493 has no significant toxic or side effects on mice, therefore it could be safely applied in disease control for agricultural production.
