为充分开发利用红千层(Callistemon rigidus R.Br.)植物资源,采用水蒸汽蒸馏法提取了红千层叶的精油(CrEO),测定了CrEO的总抗氧化能力;以人工合成抗氧化剂二丁基羟基甲苯(BHT)和丁基羟基茴香醚(BHA)作为阳性对照,测定了其对羟基(OH)自由基、1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除能力;并采用浸液法测试了CrEO对中华按蚊四龄幼虫的毒杀效果. 结果显示,CrEO的总抗氧化活性强于BHT,但弱于BHA,总抗氧化活性随浓度升高呈现显著增强的趋势. CrEO对DPPH自由基的清除率达到50%时的质量浓度(IC50)为6.452 mg/mL,对羟基自由基的IC50值小于1.0 × 10-5 mg/mL. 在毒杀实验中,CrEO对中华按蚊四龄幼虫的24 h毒杀LC50(半致死浓度)值为293.31 mg/L. 本研究结果表明,CrEO具有较强的抗氧化活性,对中华按蚊幼虫具有一定的毒杀活性,具备进一步开发成为天然的抗氧化剂和植物源杀虫剂的潜力.
Abstract:
In order to fully exploit and utilize the plant resources of Callistemon rigidus, we extracted essential oil from its leaves (CrEO) by steam distillation. The total antioxidant capacity, hydroxyl free radicals and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals scavenging of CrEO were measured using butylated hydroxytoluene (BHT) and butyl hydroxyanisole (BHA) as positive controls. Meanwhile, the larvicidal activity of CrEO against the larvae of Anopheles sinensis was evaluated by immersion method. The results indicated that CrEO had strong antioxidant ability. The total antioxidant capacity of CrEO was higher than that of BHT but lower than BHA, with the total antioxidant activity increased with the concentration. In addition, CrEO had good free radical scavenging activity. The half maximal inhibitory concentration (IC50) against DPPH radicals was 6.452 mg/mL, and the IC50 value against OH radical was less than 1.0 × 10-5 mg/mL. In addition, results of the 24 h mosquito larvicidal assay showed that CrEO had certain toxicity against larvae of A. sinensis with LC50 value of 293.310 ?g/mL. These findings suggested that CrEO can be a promising natural antioxidant and botanical mosquito control agent.
