为探讨使用诱导剂提高雷公藤(Tripterygium wilfordii Hook. f)叶片内酯醇(Triptolide,TPL)含量的可能,以同一无性系雷公藤2年扦插苗为实验材料,采用水培的方式,研究了不同浓度的水杨酸(SA)(50、5、0.5 mg L-1)、茉莉酸甲酯(MJ)(1、0.1、0.01 mmol L-1)、6-苄基腺嘌呤(6-BA)(0.15、0.1、0.05 mg L-1)和吲哚乙酸(IAA)(50、25、5 mg L-1)诱导剂在第0、10、20、30、40天对雷公藤叶片丙二醛(MDA)、渗透调节物质、主要光合色素含量和抗氧化酶活性,以及第40天对根、茎、叶TPL含量的影响. 结果表明:诱导后的叶片TPL、CAT、POD、Pro、MDA、Chl a和Chl b含量均高于对照组(P < 0.01),而SOD和SPC含量均低于对照组(P < 0.01);相关性分析显示TPL含量与SPC、SOD呈极显著负相关关系,相关系数分别为-0.69、-0.73,与CAT、POD呈极显著正相关关系,相关系数分别为0.69、0.71;主成分分析筛选确定1 mmol L-1 MJ对雷公藤TPL含量提高效果最理想,该处理叶片中Pro、POD、Chl a、Chl b含量最高,显示了其较高的渗透调节和抗氧化能力,以及较好的生长状况. 综上,与对照组相比,采用外源诱导剂处理显著增加了TPL的含量,但是幼苗对这4种诱导剂的响应和耐受机制存在差异.
Abstract:
Low available triptolide (TPL) is one of the major limitations for wide clinical applications of Tripterygium wilfordii Hook.f.. This study aimed to understand the effects of inducers on physiological and biochemical properties and TPL in leaves of T. wilfordii seedings. The experiment included treatments of four different inducers, Salicylic Acid (SA), Methyl Jasmonate (MJ), 6-Benzylaminopurine (6-BA) and Indole Acetic Acid (IAA), of different concentrations. On day 0, 10, 20, 30, 40, we analyzed the physiological and biochemical responses (including malondialdehyde (MDA), proline (Pro), soluble protein content (SPC), catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), chlorophyll a (Chl a), chlorophyll b (Chl b), carotenoids (Caro)) in leaves; we also analyzed the TPL contents in rhizome and leaves on 40 d. The influence of treatments of the 9 physiological and biochemical indexes was analyzed with single factor experiment statistical analysis, two-factor experiment statistical analysis, correlation analysis and principal component analysis. The results showed a higher content of CAT, POD, Pro, MDA, Chl a and Chl b but a lower content of SOD and SPC than the control after induction, with no obvious change in Caro. Correlation analysis indicated that TPL was negatively correlated to SPC (R2 = -0.69) and SOD (R2 = -0.73) but positively to CAT (R2 = 0.69) and POD (R2 = 0.71). Principal component analysis showed that the 1 mmol L-1 MJ solution treatment was the most effective, resulting in higher leaf contents of Pro, POD, Chl a and Chl b than others, which suggested a higher osmotic adjustment ability and antioxidant capacity, and better growth status and photosynthetic capacity in T. wilfordii seedings. These results showed that treatment with different inducers may, through different responses and tolerance mechanisms of the seedling, increase TPL contents in T. wilfordii leaves.
