摘要以龙眼胚性愈伤组织为起始材料,研究了胚性悬浮细胞培养的影响因素、继代保持方法及其体胚发生与植株再生. 结果表明,建立龙眼优良胚性悬浮细胞系的技术关键在于:由松散型胚性愈伤组织作为起始材料;以去除琼脂的保持松散型胚性愈伤组织的培养基作为启动培养基;采用分别附加5 mg L-1 AgNO3、100 mg L-1肌醇的培养基交替继代保持.胚性悬浮细胞在附加2%蔗糖、0.7%琼脂、400 mg L-1LH、100 mg L-1肌醇的MS固体培养基上形成的体胚数量可达7×104~14×104g-1.经过成熟培养后,体胚转换植株的频率在65%以上. 图版1 图1 表3 参16
Abstract:
AbstractThe factors affecting establishment of embryogenic suspensions from embryogenic calli, the methods of suspension maintenance and plant regeneration via somatic embryogenesis of longan were studied. The results showed that the key factors for establishing an excellent embryogenic suspension were as follows: initiating from friable embryogenic callus in the liquid medium with the same components as the subculture medium of friable embryogenic callus free of agar; maintaining suspensions via alternative culture of the media amended with 5 mg L-1 AgNO3and 100 mg L-1 myoinositol, respectively. The embryogenic suspension cells formed somatic embryos at the frequency of 70 000~140 000 g-1 on the solid MS medium amended with 2% sucrose, 0.7% agar, 400 mg L- 1LH and 100 mg L-1 myoinositol. The somatic embryos were converted into plantlets at the frequency of over 65% after maturation. Plate 1, Fig 1, Tab 3, Ref 6
