Cholesterol is a new kind of steroidal resources, which is composed of steroidal mother nucleus and side chain. A new strain 5901, which effectively converted cholesterol to 4-cholesten-3-one, was isolated from soil samples and identified as Rhodococcus sp. by the morphological observation and 16S rDNA gene sequence analysis. The optimal condition for the strain to convert cholesterol was 30 °C for growth and transformation, and 8.0 for initial pH. The conversion of cholesterol reached 95% under the optimal condition after 24 h when the substrate concentration was 5 g/L. From practical point of view, this strain has relatively higher substrate tolerance and transformation rate than the earlier reported strains. In addition, it also oxidizes 3b-hydroxyl group and ?5→4 isomerization of other steroids. These results suggest that it may be a useful strain for potential industrial application.
