Abstract:
Abstract The activity and conformation changes of polygalacturonaseinhibiting protein (PGIP) from wheat denatured by pH and temperature was studied by the measurement of circular dichroic (CD) spectra. The native PGIP of wheat exhibited a characteristic CD spectral negative band centered at 211 nm. It was estimated to contain about 43.7% βsheet and 13.1% αhelix. This is the first study on the secondary structure of wheat PGIP. With the change in activity of wheat PGIP induced by pH and temperature, CD spectra changed. The band was smaller when activity was weaker, and it had blue shift while wheat PGIP was inactive. The further studies showed that βsheet was directly interrelated with the changes of activity of PGIP from wheat.Fig 2, Tab 2, Ref 23
