Abstract:
The accumulation of rare earths in different genotypes of tea trees is confirmed to show a significantly different. Unfortunately, the effect of accumulation of rare earths on the physiological mechanism of tea trees is unclear. In this study, two tea tree varieties ( Shuixian and Tieguanyin) were chosen as candidates to understand the La adsorption amount, growth condition, and the changes in ultrastructure of leaf cell under various concentrations of La (0.08 mmol/L, 0.4 mmol/L and 1.2 mmol/L) by using indoor nutrient fluid cultural method. The results showed that with the increase in La concentration, the absorption amount of La decreased firstly and then increased at different leaf positions. Under the concentration of La at 1.2 mmol/L, the accumulation amount of La at different leaf positions of Shuixian species was 0.3-1.7 fold higher than that of Tieguanyin. Under low concentration of La, the growth condition was vigorous and the new shoots were in mature and health condition for both tea tree varieties. In high concentration of La treatment, the plants were shorter, yellowish-green in color with tarnish were obviously observed as well as the growth development of new shoots delayed. From the observation of scanning electron microscope, no significant difference was observed between cell structure for control sample and low-concentration La treated sample, whereas the cell wrinkles markedly emerged and wax were grown around the stomata under higher concentration of La treatment. Compared with the two varieties, Tieguanyin had less wax coverage, larger stomatal density, and less damage whereas the Tieguanyin showed stronger tolerance. Transmission electron microscope observation showed that the leaf organelles of two tea varieties were damaged and cytoplasmic components were lost in high concentration of La treatment, in particular to Shuixian species.This study showed that the narcissus species is susceptible to be poisoned by La metal and Tieguanyin showed stronger tolerance. This investigation explored the preliminary understanding regarding the physiological reactions of different tea tree varieties to rare earths, furthermore provided useful information for screening the low rare earths accumulation tea tree.
