Abstract:
Abortiporus biennis is a medicinal white-rot fungus with antioxidation and anti-tumor effects, which can degrade lignin and cause white decay of wood. In the present study, A. biennis strain WX-02 isolated from Wuxi, Jiangsu Province, was used as the material. Optimal culture conditions of mycelia, antioxidant activities, and Cu2+ induced laccase production of liquid fermentation were further investigated. Optimum culture conditions based on single factor experiments have revealed that the optimum carbon source for strain WX-02 mycelial growth was glucose, while the optimum nitrogen sources were beef extract and soybean powder. When the C/N ratio was 40/1, the mycelia demonstrates the greatest growth vigor. The optimum growth factors were determined to be corn flour and vitamin C, while the optimal pH and temperature were 7.0 and 32 ℃, respectively. With the culture conditions of a 10% (V/V) inoculation amount at 28 ℃ and 150 r/min for 72 h, the fermentation broth using PD medium manifested a total antioxidant activity of 2.76 ± 0.31 mmol/L (FeSO4) and 0.40 ± 0.01 mmol/L (Trolox), DPPH radical scavenging ratio of 100.00 ± 0.00%, and SOD activity of 15.75 ± 0.25 U/mL. Whereas the fermentation broth using the optimum culture condition medium demonstrated a total antioxidant activity of 0.20 ± 0.03 mmol/L (FeSO4) and 0.16 ± 0.01 mmol/L (Trolox), DPPH radical scavenging ratio of 59.06 ± 0.61%, and SOD activity of 15.65 ± 0.62 U/mL. Further study of Cu2+ on extracellular laccase production revealed that Cu2+ of 0.25 mmol/L can significantly enhanced the enzyme production with the highest activity of (1.82 ± 0.02) × 105 U/mL after the liquid fermentation at 28 ℃ at 150 r/min for 96 h, which was 1.72 times as much as the control group. The strain WX-02 demonstrated considerable high antioxidant and laccase activities, suggesting its potential applications in medicinal and environmental management.
