Abstract:
In order to find the effects of cadmium (Cd) stress on the secretion of low molecular weight organic acids (LMWOA) by Agaricus bisporus mycelia and the contents of Cd in the fermentation broth. The contents of Cd and 8 kinds of LMWOA, including formic acid, acetic acid, citric acid, malic acid, oxalic acid, tartaric acid, succinic acid and fumaric acid , in fermentation broth and their correlation under different Cd stress time and Cd stress concentration were analyzed by shaking flask experiment using Agaricus bisporus A15 as material. The results found: The types and contents of LMWOA in fermentation broth were significantly different with the increase of stress time and concentration (P < 0.05 ). With the prolongation of stress time, the change trends of formic acid, malic acid, succinic acid and acetic acid in the fermentation broth were the consistent. Formic acid increased basically, malic acid and succinic acid increased first and then decreased, acetic acid showed a "double peak" pattern. The change trends of oxalic acid, tartaric acid, fumaric acid and citric acid were different . Without Cd stress, oxalic acid and tartaric acid increased, citric acid and fumaric acid decreased. Under Cd stress, oxalic acid and tartaric acid showed a "double peak" pattern, while citric acid and fumaric acid increased first and then decreased. Cd concentration of Agaricus bisporus increased with the increase of Cd concentration, and increased first and then decreased with the extension of Cd stress time. The correlation analysis showed that: At 2h, 8h and 1d of Cd stress, the content of Cd in fermentation broth was negatively correlated with fumaric acid, citric acid, malic acid and acetic acid, positively correlated with succinic acid. At 3d, 5d and 7d of Cd stress, the content of Cd was negatively correlated with oxalic acid and positively correlated with malic acid. During the whole process, the content of Cd was positively correlated with tartaric acid. Under 0.50 and 1.00 mg/ Cd stress, the content of Cd was negatively correlated with oxalic acid, tartaric acid, malic acid, acetic acid, citric acid, fumaric acid and succinic acid . Under 2.00、6.00 and 15.00 mg/L Cd stress , the content of Cd in culture medium was positively correlated with oxalic acid, tartaric acid, formic acid, acetic acid, succinic acid and total acid, negatively correlated with citric acid and fumaric acid. Obviously, Cd stress could significantly affect the kinds and contents of LOWOA secreted by Agaricus bisporus mycelia , which play an important role in Cd resistance of Agaricus bisporus.
