Abstract:
This research aimed to isolate the chromium-resistant bacterial endophyte strains and to evaluate their chromium resistance and plant growth promoting traits. LB medium and two isolate approaches were employed to isolate endophytic bacteria from Deyeuxia scabrescens (Griseb.) growing in chromium contaminated soil. In order to analyze the chromium resistance of the endophytic bacteria, the isolates were inoculated on medium of different chromium concentration. To verify the chromium stress resistance and removal ability, the isolates with strong chromium resistance in preliminary screening were measured for the OD value in LB liquid medium containing different concentration of chromium. The diversity and plant growth promoting activity of the endophytes were also analyzed. Out of the 33 endophytic bacteria isolated from Deyeuxia scabrescens (Griseb.), 27 strains could grow on media with chromium concentration greater than 500 ug/mL, with 5 isolates showing chromium stress resistance up to 1000 μg/mL. Additional experimental revealed the high chromium removal rate of SCAUE9002 (79.55%) and SCAUE9006 (75.27%) in LB medium (300 mg Cr6+/L, 48 h incubation). Phylogenetic analysis based on 16S rDNA sequences showed that these isolates belonged to 6 species, including Bacillus, Staphylococcus, Terribacillus, Ochrobacterium, Ignatzschineria and Alcaligenes. In terms of plant growth promoting ability, indole-3-acetic acid (IAA) production was detected in 24 strains (72.73%), Siderophore production was positive in 18 strains (54.54%), 10 strains (30.3%) of the test isolates showed ability to solubilize phosphate, and 13 strains produced cellulases (39.40%). Our research suggested that bacterial endophytes from chromium contaminated area are promising in bioremediation for chromium contaminated soils.
