Abstract:
This study aimed to understand the effect of VC, VE and Glutamine (Gln) addition in diet on the cold stress resistance of rats. A cold-stress model was first established on 30 clean female Wistar rats randomly divided into three groups (N = 10). The experimental group was intragastricly administered a mixture of VC, VE and Gln (VC 600 mg/kg, VE 100 mg/kg and Gln 1 000 mg/kg); the cold stress group were exposed to the temperature of 11±1 oC for 24 h after model establishment; and the normal temperature group were kept under temperature of 21±1 oC. The concentration of antioxidant parameters in serum (NE, GSH-PX, SOD, T-AOC, MDA) were detected by ELISA technology. The relative expression of HSP70 mRNA was determined by method of qRT-PCR. The results indicated that: (1) the concentration of NE in serum tended to significantly increase after 24 h of cold stress (P < 0.01); (2) the concentration of antioxidants in serum tended to significantly reduce after 24 h of cold stress (P < 0.01); (3) the HSP70 mRNA level of q-PCR test of the cold stress group was extremely significantly higher than that of the normal temperature group (P < 0.01). From the above results, it could be concluded that the compound nutrients additive of VC, VE and Gln has a good effect on anti-cold stress of Wistar rats, with the optimal dose in basal diets as VC 600 mg/kg ,VE 100 mg/kg and Gln 1 000 mg/kg.
