There are many different extreme ecological environmental conditions with low organic matters in Xinjiang, China. In order to understand the microorganism communities and the resource of biodiversity of the extreme ecological environmental conditions, seven typical Xinjiang oligotrophic environment samples were investigated. In this study, an improved diluted nutrient medium method was used to isolate the cultivable microorganism, and meanwhile the total microbial DNA was extracted from those 7 samples respectively. The 16S rRNA V3 fragment was amplified by PCR, and the polymorphism of this fragment was analyzed by DGGE. Including fungus, fifty oligotrophic strains were preliminary identified from more than 800 single colonies, and some unknown bacteria were indicated by sequence comparison of the 16S rRNA (randomly selecting 10 strains of bacteria) with Genbank. A study of the bacterium resources multiplicity and the similarity from the 7 different oligotrophic environmental conditions suggested that the diversity of bacterial community was very high. This research would provide significant information for developing and using the Xinjiang oligotrophic environmental microorganism resources, and for protecting the Xinjiang oligotrophic ecological environment.
