Isolation and Identification of Fungi from Rotted Tuber of Sweet Potato

Four strains of mycelial fungi from rotted tubers of sweet potato were isolated and numbered as SP-1, SP-2, SP-3 and SP-4, respectively. All of them could grow rapidly on the tubers of sweet potato. Their morphologies of colony and structure were observed on PDA medium. The total DNA of the fungi was extracted, and then their 18S rDNA was amplified, cloned and sequenced. The lengths of the 18S rDNA of the four strains were 1 801 bp, 1 769 bp, 1 768 bp and 1 770 bp,respectively. Their sequences were aligned with the 18S rDNA of other fungi in GenBank and the highly homologous sequences obtained were used to construct phylogenetic tree. According to the results of DNA molecular identification and morphological observation of the four fungi, they were identified as Rhizopus stolonifer SP-1, Fusarium oxysporum SP-2, Hypocrea koningii SP-3, and Hypocrea sp.
