Abstract:
Protocorms of Gladiolus hortalans emerged from the segments of bud cuts when 2,4-D was added into the hasal MS medium. The influences of hormone, carbone source, temperature, illumination, pH and culture style on callus growth and protocorm formation were investigated. The proper conditions: ρ(2,4-D)=1mg/L, ρ(NAA)=0.5mg/L, ρ(Sugar)=30g/L, 0=21-26℃, pH 5.8 and shallow liquid medium illumination culture. Under the optimum conditions, a self-made double tower-plate radius-flow bioreactor was used for the protocorm culture. The results showed the protocorms grew well in the bioteactor. After the tissue was cultured for 28d, fresh weight was increased by 8 times and number of protocorms by 9 times.
