Abstract:
Abstract A16, an engineering strain with a high ability of producing alkaline protease was obtained by fusing protoplasts of Bacillus licheniformis 2709 and Bacillus subtilis BD105〔pDW2 (pMk4 and alkaline protease gene)〕. In situ hybridization data indicated that A16 carried genes from BD105 (pDW2) and B.licheniformis 2709. Cell proliferation aspect and phenotype of A16 was similar to that of 2709, and its activities of fermentation were 50%~100%, higher than those of 2709 under the same conditions. The maximum activities of protease were 30 000 u/mL from A16 in the optimum fermentation. The optimal catalyzing temperature and pH for alkaline protease of A16 were also similar to those of 2709 and they could be used for industry production. Fig 3, Tab 5, Ref 22
