Abstract:
Abstract 26 varieties of common wheat (Triticum aestivum L.) including 15 Chuanyu varieties and their 11 parents, whose HMW-GS Glu-D1 locus was known, were investigated using PCR technique. According to the gene sequences of Dx5 and Dy10, two pairs of primers were designed to detect these two genes respectively. Dx5 gene was detected with regular PCR and Dy10 gene was detected with PCR- based AS-PCR. The results showed that both Dx5 and Dy10 genes could amplify specific fragments and the materials which with Dx5 genes might also possess Dy10 genes. Therefore, the high quality 5+10 subunits can be effectively and rapidly identified by the molecular primers designed.
